Functional macrophage modulation via neutrophils in inflammatory arthritis
Neutrophils and macrophages play a central role in the pathogenesis of inflammatory arthritis. However, the spatiotemporal dynamics of neutrophil migration into joints and their interaction with tissue resident cells, particularly macrophages, remain poorly understood. We uncovered that neutrophils can infiltrate joints through different anatomical routes and exhibit transcriptional heterogeneity in inflamed joints. Different neutrophil states show unique spatial interaction patterns with tissue-resident macrophage subsets. Thereby, neutrophils are uniquely positioned to tip the balance to either promote a proinflammatory or a resolving macrophage state. Accordingly, we hypothesize that modulating neutrophil states can balance the pro-inflammatory and pro-resolving abilities of synovial macrophages to promote homeostasis. To address these interactions, we elucidate the influence of different migration routes, the developmental origin of different neutrophil states and their influence on macrophage states. Furthermore, we also explore the targeted modulation of neutrophil-macrophage interactions to restore homeostasis in inflammatory arthritis.
