Regulatory role of type I interferon signalling in skewing anti-tumoural granulopoiesis

Current cancer immunotherapies tend to neglect the critical role of innate immunity. Yet, understanding how neutrophils acquire tumour- promoting functions and how to counteract them is thus of great interest. We have previously reported that type I interferon (IFN) signalling enhances anti-tumourigenic properties of neutrophils. Our data from the first funding period suggest that this type I IFN-driven reprogramming may be initiated early during granulopoiesis. Notably, in cancer, downregulation of IFN signalling leads to tumour progression, suggesting that tumours may hijack granulopoiesis by actively dampening this pathway to skew neutrophil development toward pro-tumoural phenotypes. Building on these insights, we aim to investigate how type I IFN shapes neutrophil functional states in cancer by acting on early stages of granulopoiesis.
